    Simulation analysis of crack cause of concrete overflow dam for Hadashan Hydro Project by 3-D FEM

    AbstractThis paper is mainly to study the cracking reasons for concrete overflow dam of Hadashan Hydro Project. The three-dimensional finite element method (3-D FEM) is developed to simulation analysis the temperature and thermal stress distribution in the concrete overflow dam during the construction period. The results show that the crack of the concrete overflow dam is temperature crack, mainly due to the combined action of the internal thermal gradient and the external restraints; and dramatic changes in ambient temperature exacerbate cracking of early-age concrete. Finally, the results are applied to provide some references for the construction in the related fields

    Indole-thiophene conjugate inhibits proliferation of human cervical cancer cell lines through DNA damage

    Purpose: To investigate the inhibitory effect of indole-thiophene conjugate (ITC) against cervical cancer cells. Methods: The effect of ITC on the proliferation of cervical cells was determined using 3 (4,5 dimethylthiazol 2 yl) 2,5 diphenyltetrazolium bromide (MTT) assay. The apoptosis-inducing effect of ITC was analysed with flow cytometry, while its effect on cell invasion was assessed using Transwell assay. Results: ITC inhibited proliferation of HeLa and Caski cancer cell lines, but it had no cytotoxicity against HCvEpC normal epithelial cells. Exposure to ITC at a dose of 12 μmol/L reduced the viability of HeLa and Caski cells to 22.56 and 24.78 %, respectively (p < 0.05). ITC treatment of HeLa cells enhanced the proportion of apoptotic cells. Exposure to ITC at a dose of 12 μmol/L led to near-complete inhibition of the invasive potential of HeLa cells. Moreover, exposure of HeLa cells to ITC downregulated the protein expressions of MMP-2 and MMP-9 (p < 0.05). The expressions of Bcl-2, p-ERK1/2 and p-Akt were markedly decreased in HeLa cells by ITC exposure. In addition, ITC increased Bax expression, and decreased Bcl-2/Bax ratio (p < 0.05). Conclusion: ICT inhibits the proliferation and invasion of cervical cancer cells, and induces their apoptosis. It exhibits these effects via the suppression of Akt and ERK phosphorylation, thereby downregulating the PI3K and MAPK pathways. Therefore, ITC may be beneficial for the treatment of cervical cancer

    MiR-429 suppresses the progression and metastasis of osteosarcoma by targeting ZEB1

    MiR-429 functions as a tumor suppressor and has been observed in multiple types of cancer, but the effects and mechanisms of miR-429 in osteosarcoma are poorly understood. This study is performed to evaluate the functions of miR-429 in the progression of osteosarcoma. Firstly, the miR-429 expression in osteosarcoma tissues and oste- osarcoma cells was detected using real time PCR, and the relationship between miR-429 expression and overall survival of osteosarcoma was analyzed. Secondly, the effects of miR-429 on the migration, invasion, proliferation and apoptosis of osteosarcoma cells were evaluated using transwell assay, wound-healing assay, CCK-8 assay and flow cytometry, respectively. Proteins related to epithelial-mesenchymal transition (EMT), E-cadherin, Vimentin, N-cadherin and Snail, were also detected using Western blot. Finally, the target gene of miR-429 in osteosarcoma was predicted and verified using dual luciferase assay and the expression correlation between them was analyzed using Pearson’s correlation. MiR-429 was down-regulated in osteosarcoma tissues and osteosarcoma cells; the expression level of miR-429 was associated with the prognosis of osteosarcoma. High level of miR-429 in osteo- sarcoma cells significantly suppressed the migration, invasion and proliferation of cells but induced cells apopto- sis. Furthermore, high level of miR-429 in osteosarcoma cells obviously increased the expression of E-cadherin protein but decreased the expression of Vimentin, N-Cadherin and Snail proteins. EMT inducer ZEB1 was the target gene of miR-429 and the expression of ZEB1 was negatively related to the miR-429 expression in osteosar- coma. In conclusion, miR-429 may functions as a tumor suppressor and be down-regulated in osteosarcoma. MiR- 429 may suppress the progression and metastasis of osteosarcoma by down-regulating the ZEB1 expression

    Dynamic modeling and control of a novel XY positioning stage for semiconductor packaging

    This paper presents the dynamic modeling and controller design of an XY positioning stage for semiconductor packaging. The XY stage is directly driven by two linear voice coil motors, and motion decoupling between the X and Y axes is realized through a novel flexible decoupling mechanism based on flexure hinges and preloaded spring. Through bond graph method, the dynamic models of X- and Y-axes servomechanisms are established, respectively, and the state space equations are derived. A control methodology is proposed based on force compensations and the performance of the XY stage is investigated by simulations and experimental tests. The results show that the XY stage has good performance. When the reference displacements are defined as 2 mm, the settling time of the X-axis movement is 64 ms, and the overshoot is 0.7%. Y-axis settling time is 62 ms, and the overshoot is 0.8%. X-axis positioning accuracy is 1.85 μm and the repeatability is 0.95 μm. Y-axis positioning accuracy and repeatability are 1.75 μm and 0.9 μm, respectively. In addition, the stage can track linear, circular and complex trajectories very well

    CT-guided 125I brachytherapy combined with chemotherapy for the treatment of unresectable or locally advanced pancreatic carcinoma

    PURPOSEWe aimed to explore the feasibility and clinical effectiveness of percutaneous CT-guided iodine-125 (¹²⁵I) brachytherapy combined with chemotherapy for the treatment of patients with unresectable or locally advanced pancreatic carcinoma (PC).METHODSWe retrospectively reviewed 66 patients with Stage III and IV PC who had received chemotherapy. A total of 35 (53%) patients receiving 125I brachytherapy and chemotherapy (gemcitabine + cisplatin, GP) were classified as Group A, and 31 (47%) patients who received GP chemotherapy alone were categorized as Group B. The evaluated indications were local control rate (LCR), local progression-free survival (LPFS), overall survival (OS), treatment-related complications, and the degree of symptom relief. Kaplan-Meier curves, log-rank test and Cox regression models were generated and used for further analysis to identify predictors of outcomes.RESULTSThe median follow-up time was 6.00±0.84 months. The 1-, 3-, 6-, 12- and 18-month LCRs for Group A were 100% (35/35), 89.3% (25/28), 71.4% (15/21), 37.5% (3/8) and 33.3% (1/3), respectively; and those for Group B were 87.1% (27/31), 69.6% (16/23), 41.2% (7/17), 14.3% (1/7) and 0% (0/3), respectively. The LCR differed at 1-, 3- and 6-months (P = 0.032; P = 0.009; P = 0.030; respectively). The median LPFS was 7.00±0.30 months and 5.00±0.75 months for Groups A and B (P = 0.023), respectively; however, the median OS of the groups were not significantly different (8.00±0.77 months vs. 6.00±1.04 months. P = 0.917). No life-threatening complications occurred during or after the procedures. Patients in Group A experienced better pain control and relief of abdominal distension than those in Group B.CONCLUSIONCT-guided 125I brachytherapy is a feasible, safe, and valuable treatment for patients with unresectable PC
